Induced pluripotent stem cell-derived CD8 alpha-beta T cells (glypican-3-targeted) is an investigational cell therapy developed by Century Therapeutics. It consists of CD8αβ T cells differentiated from induced pluripotent stem cells (iPSCs). These cells are engineered to express a T-cell receptor (TCR) targeting glypican-3 (GPC3) peptides presented by HLA-A24 on tumor cells. Additionally, they possess innate killing capabilities mediated by receptors such as Killer cell lectin-like receptor K1 (NKG2D) and CD226 (DNAM1). The cells are further armored with constitutive secretion of Interleukin-15 (IL-15) and Interleukin-21 (IL-21) to enhance their persistence and serial killing activity, and include immune evasion edits to prevent rejection. This therapy is being developed for the treatment of liver cancer and lung cancer, with administration via intravenous or intraportal routes.
