Induced pluripotent stem cell-derived natural killer cells expressing soluble interleukin-15 (iPS-NK-IL15) is an engineered cell therapy consisting of natural killer (NK) cells derived from induced pluripotent stem cells (iPSCs) and genetically modified to secrete soluble interleukin-15 (IL-15). This approach is designed to overcome the immunosuppressive tumor microenvironment, particularly in the central nervous system (CNS), where low levels of endogenous IL-15 typically limit NK cell survival, maturation, and effector function. By providing an autocrine and paracrine source of IL-15, these cells maintain high cytotoxic activity and stimulate the expansion of effector Type 1 innate lymphoid cells. This candidate is a derivative of the iPSC-NK platform (such as FT538) developed by Fate Therapeutics and is being investigated for the treatment of breast cancer brain metastasis (BCBM) and other malignancies.
