Overview

Induced pluripotent stem cell-derived natural killer T cells (iPSC-iNKT cells) are an allogeneic cellular immunotherapy consisting of invariant natural killer T (iNKT) cells generated from induced pluripotent stem cells (iPSCs). These cells are designed to bridge innate and adaptive immunity by recognizing glycolipid antigens, such as alpha-galactosylceramide (α-GalCer), presented by CD1d molecules. Upon activation, iPSC-iNKT cells produce high levels of interferon-gamma (IFN-γ), which activates other immune cells like NK cells and CD8+ T cells to exert anti-tumor effects. The therapy is being developed primarily by RIKEN and the Japan Agency for Medical Research and Development (AMED) for the treatment of recurrent or advanced head and neck squamous cell carcinoma. In clinical trials, they are often administered in combination with α-GalCer-pulsed dendritic cells to enhance their activation and therapeutic efficacy.
