Overview

Indusatumab vedotin is an antibody-drug conjugate (ADC) developed for the treatment of pancreatic cancer and other gastrointestinal cancers. It consists of a fully human monoclonal antibody (indusatumab) targeting guanylate cyclase 2C (GUCY2C), a receptor overexpressed in certain gastrointestinal tumors, conjugated to the cytotoxic agent monomethyl auristatin E (MMAE). Upon binding to GUCY2C-expressing tumor cells, the ADC is internalized and MMAE is released intracellularly, where it inhibits tubulin polymerization, causing cell cycle arrest at G2/M phase and inducing apoptosis. The drug was in clinical development but its trials were terminated due to insufficient efficacy[1][3][4][5][7].
