INE963 is a potent, fast-acting small molecule antimalarial agent from the 5-aryl-2-amino-imidazothiadiazole (ITD) class. It was discovered by the Novartis Institute for Tropical Diseases and further developed in partnership with Medicines for Malaria Venture (MMV). INE963 demonstrates high potency against blood-stage Plasmodium falciparum, with EC50 values in the low nanomolar range. It exhibits rapid parasite clearance kinetics similar to artemisinin and has shown curative potential in preclinical models after a single oral dose. The compound features a high barrier to resistance, no cross-resistance with existing antimalarials, and a long predicted human half-life (~60 hours), supporting its potential as a single-dose cure for uncomplicated malaria.
