Overview

Inetetamab + Cyclophosphamide is a drug combination being investigated for the treatment of HER2-positive breast cancer. Inetetamab is a novel recombinant humanized anti-HER2 monoclonal antibody. It shares the Fab domain with trastuzumab but has a modified Fc domain with alterations at amino acid positions 359 (D359) and 361 (L361). This trastuzumab analog possesses the exact F(ab')2 of trastuzumab fusing into a different IgG1 Fc allotype. It has demonstrated comparable bioactivity to trastuzumab, including binding affinity for the HER2 antigen, protein folding, thermal stability, and the inhibition of tumor cell growth. It is being explored as a treatment option for patients who have developed resistance to trastuzumab. Cyclophosphamide is a chemotherapeutic agent belonging to the alkylating class. It works by interfering with DNA replication in rapidly dividing cells. The combination of inetetamab and cyclophosphamide has shown distinct antitumor effects in xenograft mouse models.
