The combination of inetetamab, pyrotinib, and nab-paclitaxel is a triple-drug neoadjuvant regimen being investigated for the treatment of HER2-positive breast cancer. **Inetetamab** is a recombinant humanized anti-HER2 monoclonal antibody that targets the extracellular domain of the HER2 receptor. **Pyrotinib** is an orally active, irreversible pan-HER tyrosine kinase inhibitor (TKI) that targets the intracellular kinase domains of HER1 (EGFR), HER2, and HER4. **Nab-paclitaxel** (nanoparticle albumin-bound paclitaxel) is a taxane-based chemotherapy agent that inhibits microtubule dynamics. This regimen utilizes a dual HER2 blockade strategy (extracellular and intracellular) combined with cytotoxic chemotherapy to maximize pathological complete response (pCR) rates in patients with early or locally advanced HER2-positive breast cancer. It is currently being evaluated in Phase II clinical trials sponsored by institutions such as Xijing Hospital.
