Overview

Combination therapy with infliximab and azathioprine is used primarily for the treatment of moderate-to-severe Crohn's disease. Infliximab is a chimeric monoclonal IgG1 antibody that targets tumor necrosis factor alpha (TNF-α), a key proinflammatory cytokine, thereby disrupting its interaction with TNF receptors and downregulating inflammatory pathways. Azathioprine is an immunosuppressive agent that inhibits purine synthesis, leading to reduced proliferation of B and T lymphocytes and modulation of immune responses. Clinical trials have shown that this combination provides superior rates of corticosteroid-free clinical remission and mucosal healing compared to either drug alone, though it carries increased risks such as infection or lymphoma due to combined immunosuppression[1][2][4][5][7].
