Influenza A (H5N1) inactivated split-virion vaccines are non-infectious vaccines developed for active immunization against the H5N1 subtype of the influenza A virus. These vaccines contain purified hemagglutinin antigen derived from an H5N1 strain that has been propagated in embryonated hen’s eggs and then chemically inactivated. The "split-virion" process involves disrupting the viral envelope to reduce reactogenicity while preserving immunogenicity. Most formulations are adjuvanted with oil-in-water emulsions such as AS03 or MF59 to enhance immune response and allow for dose sparing. The primary mechanism is induction of neutralizing antibodies targeting the hemagglutinin protein on the surface of H5N1 viruses, thereby preventing infection. These vaccines have been developed by major manufacturers like GSK and Sanofi Pasteur and are approved or stockpiled for use during potential pandemics caused by avian-origin H5N1 strains[2][3][6].
