H9N2 whole virus refers to a vaccine modality utilizing the entire inactivated H9N2 influenza A virus to elicit an immune response. H9N2 is a subtype of avian influenza virus that is endemic in poultry across Asia, the Middle East, and Africa, and is considered a significant pandemic threat due to its ability to infect humans and provide internal genes to other highly pathogenic strains like H5N1 and H7N9. Whole virus inactivated vaccines (WIV) are typically produced by growing the virus in embryonated chicken eggs or cell culture, followed by inactivation with agents such as formaldehyde or beta-propiolactone. These vaccines contain all structural proteins of the virus, including hemagglutinin (HA), neuraminidase (NA), nucleoprotein (NP), and matrix proteins, which may provide broader cross-reactivity compared to split-virus or subunit vaccines. Clinical development has been pursued by various organizations, including the NIAID and several Chinese manufacturers, to prepare for potential zoonotic transmission and pandemic outbreaks.
