The **influenza saRNA vaccine** is an investigational self-amplifying ribonucleic acid (saRNA)-based vaccine platform designed to prevent influenza virus infections. Unlike conventional mRNA vaccines, saRNA vaccines incorporate genetic machinery (typically derived from alphaviruses) that enables the RNA to replicate itself inside host cells, leading to sustained antigen expression at much lower initial doses. In clinical development, Pfizer (in collaboration with BioNTech) and hVIVO Services Ltd have evaluated monovalent and bivalent saRNA vaccine formulations encoding the influenza A (H1N1) hemagglutinin (HA) surface antigen. Specifically, a Phase II randomized, controlled human challenge study (PIR-CSV-001) conducted by hVIVO and funded by Pfizer UK evaluated these saRNA candidates in healthy adults, assessing their efficacy and safety against a licensed quadrivalent influenza vaccine (QIV) and placebo following a quarantine-based viral challenge with the A/Delaware/55/2019 (H1N1) strain.
