Overview

A/California/7/2009 (H1N1)v-like vaccine adjuvanted with AS03 is an inactivated, split-virion pandemic influenza vaccine developed by GlaxoSmithKline (GSK) Biologicals. It was marketed under the brand names Pandemrix (manufactured in Dresden, Germany) and Arepanrix (manufactured in Quebec, Canada). The vaccine contains inactivated split-virion influenza A haemagglutinin (HA) antigen derived from the A/California/7/2009 (H1N1)v-like strain, combined with GSK's proprietary AS03 Adjuvant System—an oil-in-water emulsion containing squalene, DL-α-tocopherol (Vitamin E), and polysorbate 80. The AS03 adjuvant enhances the immunogenicity of the HA antigen, allowing for antigen-sparing (using a lower dose of antigen while achieving comparable immune responses). It is indicated for active immunization against the 2009 pandemic influenza A (H1N1) strain. Pandemrix was widely used during the 2009 H1N1 pandemic but was later associated with an increased risk of narcolepsy, leading to its discontinuation.
