Novavax's influenza VLP vaccine is a recombinant vaccine candidate developed to provide protection against seasonal influenza. The vaccine utilizes virus-like particle (VLP) technology, where the hemagglutinin (HA), neuraminidase (NA), and matrix 1 (M1) proteins of the influenza virus are co-expressed to self-assemble into particles that structurally resemble the native virus but lack genetic material. These VLPs are typically produced in Sf9 insect cells using a baculovirus expression system. This trivalent formulation was designed to elicit a broad and potent immune response, including high titers of hemagglutination inhibition (HAI) antibodies and neuraminidase inhibition (NAI) antibodies, as well as T-cell responses. Although it showed promise in Phase 1 and Phase 2 trials, particularly in older adults, Novavax eventually transitioned its influenza program focus toward its 'NanoFlu' candidate, which utilizes recombinant HA nanoparticles with the Matrix-M adjuvant.
