Overview

INGN 007 (also known as VRX-007) is a replication-competent oncolytic adenovirus type 5 (Ad5) vector designed for the treatment of various solid tumors. The vector is genetically engineered with a deletion of the early three (E3) transcription unit, which normally functions to evade the host immune system, and its replacement with the gene for the Adenovirus Death Protein (ADP, also known as E3-11.6K). Overexpression of ADP, which is typically produced late in the viral life cycle, significantly enhances the efficiency of cell lysis and the subsequent release and spread of the virus within the tumor mass. Unlike some other oncolytic adenoviruses, INGN 007 retains a wild-type E1A gene, allowing for robust replication. Originally developed by VirRx and later advanced by Introgen Therapeutics, INGN 007 demonstrated preclinical efficacy in lung, liver, and prostate cancer models and received FDA clearance for Phase I clinical evaluation.
