INGN-011 (Ad-mda7) is an investigational gene therapy developed by Introgen Therapeutics, utilizing a replication-deficient adenovirus type 5 (Ad5) vector to deliver the melanoma differentiation-associated gene-7 (mda-7), also known as Interleukin-24 (IL-24). The therapeutic approach relies on the selective pro-apoptotic activity of the MDA-7/IL-24 protein in cancer cells. Upon delivery, the transgene induces high levels of IL-24 expression, which triggers apoptosis through the induction of endoplasmic reticulum (ER) stress and the activation of the extrinsic and intrinsic apoptotic pathways, while leaving normal cells unaffected. Additionally, IL-24 acts as a cytokine that can inhibit angiogenesis and modulate the immune system to enhance anti-tumor activity. INGN-011 was evaluated in Phase 1 and Phase 2 clinical trials for various malignancies, including melanoma and non-small cell lung cancer, but development ceased following the developer's bankruptcy in 2008.
