INGN-234 (Ad-mda7) is an investigational gene therapy developed by Introgen Therapeutics, utilizing a replication-deficient type 5 adenoviral vector to deliver the melanoma differentiation-associated gene-7 (mda-7), also known as Interleukin-24 (IL-24). The therapeutic rationale is based on the ability of IL-24 to selectively induce apoptosis in a wide range of cancer cells while remaining non-toxic to normal cells. Mechanistically, the expressed IL-24 protein triggers the extrinsic and intrinsic apoptotic pathways, induces endoplasmic reticulum stress, and inhibits angiogenesis. Additionally, secreted IL-24 can exert bystander effects on neighboring tumor cells and stimulate a systemic anti-tumor immune response. Although it showed promise in Phase 1 and Phase 2 clinical trials for advanced solid tumors and lung cancer, development was halted following the bankruptcy of Introgen Therapeutics in 2008.
