Overview

INHBE-siRNA is a small interfering RNA (siRNA) therapeutic designed to silence the expression of the *INHBE* gene, which encodes the inhibin subunit beta E (also known as activin E). Genetic evidence from human populations indicates that heterozygous carriers of loss-of-function mutations in *INHBE* exhibit reduced abdominal fat and a lower risk of type 2 diabetes and coronary artery disease. The siRNA targets *INHBE* mRNA, primarily in the liver, leading to a sustained reduction in circulating activin E levels. Early clinical data from Phase 1 trials suggest that a single dose can effectively suppress activin E for up to three months, resulting in improved body composition and weight loss without the muscle mass loss often associated with other anti-obesity medications. Preclinical studies in humanized mouse models have demonstrated weight loss efficacy comparable to GLP-1 receptor agonists like semaglutide, with significant reductions in fat mass and preservation of lean muscle.
