The Inhibigen vaccine is an experimental cancer immunotherapy concept developed by Genocea Biosciences using their proprietary ATLAS™ platform. Unlike traditional neoantigen vaccines that target stimulatory mutations to activate the immune system, this approach focuses on "Inhibigens"—tumor mutations identified as actively suppressing anti-tumor T cell responses. In preclinical models, such as B16F10 melanoma, the administration of Inhibigens was shown to promote a "cold" tumor microenvironment, characterized by dramatic decreases in infiltrating T cells and myeloid cells. These Inhibigen-specific responses appear to involve non-classical inhibitory mechanisms that can overpower standard-of-care immunotherapies like anti-PD-1. The discovery of Inhibigens led Genocea to use the ATLAS platform to identify and exclude these suppressive sequences from their clinical-stage personalized cancer vaccines, such as GEN-009 and GEN-011, to ensure optimal stimulatory activity.
