Overview

Inhibitor 13 is a small molecule inhibitor of the Eleven-Nineteen-Leukemia (ENL) YEATS domain, identified by researchers at Texas A&M University. It was discovered using a NanoBRET system designed to evaluate the cellular permeability and potency of ENL inhibitors. The compound exhibits high metabolic stability and potent antiproliferative effects against mixed-lineage leukemia (MLL)-fusion cell lines. In preclinical studies, inhibitor 13 demonstrated significant survival benefits in acute myeloid leukemia (AML) xenografted mice. It served as the structural basis for the development of the optimized candidate SR-C-107 (R).
