INI-2002 is a synthetic toll-like receptor 4 (TLR4) agonist being developed as a vaccine adjuvant to enhance immunogenicity and bias responses toward Th1/Th17 profiles. It activates human TLR4 with dose-responsive NF-κB signaling in reporter cells and signals through both MyD88- and TRIF-dependent pathways in human PBMCs, inducing cytokines such as TNFα, IL-6, IP-10/CXCL10, and IL-1β. In comparative studies with the TLR7/8 agonist INI-4001, INI-2002 increased antibody responses in some vaccine contexts (e.g., SARS-CoV-2 spike/RBD emulsions) but generally showed less enhancement of functional protection than INI-4001 in anti-fentanyl and POWV vaccine models. INI-2002 is supplied and studied by Inimmune in multiple preclinical vaccine formulations and combinations with alum and/or INI-4001.[1][2][3][5][7]
