Overview

INI-4001 + atezolizumab is an investigational combination therapy for advanced solid tumors, currently being evaluated in a Phase 1 clinical trial. INI-4001 is a novel, nanoparticle-formulated agonist of Toll-like receptor 7 and 8 (TLR7/8), designed to stimulate innate immune signaling, specifically inducing production of IFNα and activating antigen-presenting cells, which in turn activates T cells. Atezolizumab is a humanized IgG1 monoclonal antibody targeting programmed death-ligand 1 (PD-L1), functioning as a checkpoint inhibitor to restore anti-tumor immune responses. The combination aims to enhance both innate and adaptive anti-tumor immunity, especially in tumor types where single-agent checkpoint inhibition is insufficient. INI-4001 is being developed by Inimmune, and the Phase 1 trial includes a cohort evaluating the combination of INI-4001 and anti-PD-L1 therapy (atezolizumab) in patients with advanced solid malignancies who have progressed or have stable disease after initial INI-4001 monotherapy.
