Overview

**INI-4001 + durvalumab** is an investigational combination therapy currently in Phase 1 clinical trials for advanced solid tumors. INI-4001 is a proprietary nanoparticle-formulated **TLR7/8 agonist** designed to activate innate immunity by stimulating Toll-like receptor 7 and 8, leading to cytokine production (notably IFNα), enhanced antigen presentation, and downstream T-cell activation. Durvalumab is a **human monoclonal antibody checkpoint inhibitor** targeting PD-L1, which blocks the PD-1/PD-L1 pathway to prevent immune evasion by tumor cells. The therapeutic rationale for combining these agents is that INI-4001 promotes immune activation, which may synergize with durvalumab's checkpoint blockade to improve anti-tumor immune responses, especially in patients not fully responsive to checkpoint inhibitor therapy alone[1][2][6].
