iNK012 is an off-the-shelf chimeric antigen receptor (CAR)-engineered natural killer (NK) cell therapy derived from induced pluripotent stem cells (iPSCs). Developed by Cell Inspire Biotechnology, the therapy is specifically engineered to target Trophoblast cell-surface antigen 2 (TROP2), a glycoprotein that is highly expressed in a variety of solid tumors and associated with poor prognosis. To overcome the challenges of the immunosuppressive tumor microenvironment and enhance the persistence and anti-tumor activity of the NK cells, iNK012 is further modified to secrete Interleukin-15 (IL-15). This autocrine or paracrine cytokine support is intended to promote the survival and expansion of the CAR-iNK cells without the need for exogenous cytokine administration. iNK012 is currently in preclinical development, with ongoing validation in animal models to assess its pharmacokinetic and pharmacodynamic profiles.
