INK713 is a potent and selective small molecule inhibitor of the delta (δ) and gamma (γ) isoforms of phosphoinositide 3-kinase (PI3K). Developed by Intellikine, the compound was investigated for its potential in treating B-cell malignancies, including various types of lymphoma and multiple myeloma. Preclinical data presented at the AACR 2010 meeting indicated that INK713 possesses high affinity for PI3Kδ and PI3Kγ (IC50 < 10 nM) with significant selectivity over the alpha and beta isoforms. Its mechanism of action involves the disruption of PI3K-mediated signaling pathways that drive the proliferation and survival of malignant B-cells.
