Overview

This is a combination adoptive cell therapy involving the infusion of autologous invariant natural killer T (iNKT) cells together with CD8+ T cells, sometimes specifically PD-1 positive CD8+ T cells. iNKT cells are a unique subset of lymphocytes that bridge innate and adaptive immunity by recognizing glycolipid antigens presented by the monomorphic molecule CD1d. They can directly kill tumor and immunosuppressive myeloid populations, secrete both Th1 and Th2 cytokines, modulate the tumor microenvironment to favor anti-tumor responses, and help prevent graft-versus-host disease in allogeneic settings[1][4][5][6]. When combined with cytotoxic CD8+ T lymphocytes—key effectors in adaptive anti-tumor immunity—the therapy aims to enhance direct tumor killing (by both cell types), promote dendritic cell maturation for improved antigen presentation, induce memory responses, and overcome immunosuppression within tumors[3][5]. In clinical studies (notably in advanced pancreatic cancer), this combination has shown safety with mostly mild adverse events (fever, flu-like symptoms) and some evidence of prolonged survival or partial response[2]. The approach is under investigation as an experimental immunotherapy for solid tumors.
