Overview

**inlexisertib + binimetinib** is an investigational oral combination therapy composed of inlexisertib (DCC-3116), an ULK kinase-1/2 inhibitor, and binimetinib, a reversible inhibitor of MEK1 and MEK2. The combination is being studied for the treatment of advanced solid tumors, particularly those with RAS/MAPK pathway dysregulation. Inlexisertib inhibits ULK kinase activity, blocking autophagy initiation—a process often upregulated in cancer cells to promote survival under stress. Binimetinib inhibits MEK1/2, cellular kinases that drive proliferation in cancer through the MAPK pathway. Preclinical data suggest the combination may overcome resistance mechanisms, decrease tumor cell proliferation, and increase antitumor efficacy when used together. The combination is administered orally, with dosing established in ongoing phase 1/2 studies[4][5][7].
