Overview

INNA-051 is a novel, first-in-class small molecule developed by ENA Respiratory as an intranasal prophylactic therapy to reduce the impact of viral respiratory infections, particularly in at-risk populations such as the elderly and those with chronic medical conditions. It acts as a potent agonist of toll-like receptor 2 (TLR2) and toll-like receptor 6 (TLR6), which are cell-surface receptors expressed by human nasal epithelial and innate immune cells. By activating TLR2/6, INNA-051 boosts the body's natural antiviral defenses at the primary site of infection—the nasal mucosa—leading to accelerated viral clearance and stimulation of antiviral host defense pathways, including type I and III interferon responses. The drug is designed for once-weekly self-administration as a dry powder nasal spray, offering fast onset (<24 hours) and durable protection against multiple respiratory viruses such as SARS-CoV-2, influenza (H1N1/H3N2), and rhinovirus. Clinical studies have demonstrated its safety profile and efficacy in both preclinical models and human trials[2][4][5][6][7].
