Overview

Innate donor lymphocyte infusion (iDLI) is an adoptive cell therapy product consisting of donor-derived innate lymphocytes, specifically enriched for natural killer (NK) cells and γδ T cells. It is developed for administration after allogeneic hematopoietic stem cell transplantation (HSCT) to treat or prevent relapse in patients with hematological malignancies such as acute myeloid leukemia (AML) and myelodysplastic syndrome (MDS). The product is manufactured via an automated immunomagnetic depletion process (e.g., using the CliniMACS Prodigy system) to remove αβ T cells and CD19+ B-cells from a donor leukapheresis product. By removing αβ T cells, iDLI significantly reduces the risk of graft-versus-host disease (GVHD) while maintaining the graft-versus-leukemia (GVL) effect mediated by NK and γδ T cells through HLA-independent recognition of malignant cells.
