Overview

INNO-235 is a preclinical, orally bioavailable, cereblon (CRBN)-based molecular glue degrader designed to concurrently induce proteasomal degradation of G1 to S phase transition 1 (GSPT1, also known as eRF3a) and casein kinase 1 alpha (CK1α), leading to potent antitumor activity in MYC-driven and TP53-mutant lymphomas, including Burkitt lymphoma.[3][5] By recruiting GSPT1 and CK1α to the CRBN E3 ubiquitin ligase complex, INNO-235 triggers integrated stress response (ISR) activation and both p53-dependent and p53-independent apoptosis, resulting in strong tumor growth inhibition in TP53-mutated Burkitt lymphoma models and offering a potential therapeutic strategy for MYC-driven malignancies with limited treatment options.[1][3][5]
