Overview

**INO-1400 + INO-1401** is a combination of two synthetic DNA plasmids encoding modified variants of human telomerase reverse transcriptase (hTERT), a tumor-associated antigen overexpressed in over 90% of cancers. Developed by **Inovio Pharmaceuticals**, these plasmids are delivered intramuscularly via the CELLECTRA electroporation device, often with or without the IL-12-encoding plasmid INO-9012 as an immune adjuvant, to induce potent T-cell responses against native hTERT. INO-1400 contains two point mutations for immune tolerance breaking while retaining >99% homology to native hTERT; INO-1401 has ~95% similarity for enhanced immunogenicity. Evaluated in a Phase 1 trial (NCT02960594) across solid tumors (e.g., pancreatic, breast, lung), it demonstrated safety, tolerability, and robust hTERT-specific CD4+ and CD8+ T-cell activation, including IFN-γ production and cytotoxic T lymphocytes expressing perforin/granzymes, with 54.8% disease-free survival at 18 months overall and correlation of activated CD8+ T cells with survival in pancreatic cancer.[1][3]
