**INO-1400 + INO-9012** is a combination DNA immunotherapy vaccine developed for the treatment of solid tumors and certain cancers at high risk of relapse. INO-1400 encodes a modified human telomerase reverse transcriptase (hTERT), an enzyme highly expressed in cancer cells but not most normal cells, prompting an immune response against cells presenting hTERT. INO-9012 encodes human interleukin-12 (IL-12), an immune activator that enhances T-cell and NK cell activity, boosting the immune response initiated by INO-1400. This combination is delivered intramuscularly followed by electroporation to facilitate cellular uptake of the plasmids. The therapy induces robust antigen-specific CD4+ and CD8+ T cell responses, resulting in immune-mediated recognition and destruction of cancer cells[1][3][4][6][7][8].
