Overview

INO-1401 is a synthetic DNA vaccine encoding a modified human telomerase reverse transcriptase (hTERT) protein. Upon intramuscular administration, typically in combination with electroporation, the encoded TERT protein is expressed and stimulates the immune system to mount a cytotoxic T-cell response against telomerase-expressing tumor cells, potentially resulting in tumor cell death. The INO-1401 plasmid encodes a further modified hTERT sequence with approximately 95% similarity to native hTERT, designed to enhance immunogenicity and break immune tolerance. It is being developed primarily for the treatment of various solid tumors at high risk of relapse[1][2][3][6].
