Overview

INO-4201 is a DNA-based vaccine developed to prevent infection by the Zaire Ebola virus (ZEBOV). It encodes a synthetic consensus antigen for the ZEBOV glycoprotein (GP), designed to provide broad immune coverage against diverse outbreak strains of the virus. The vaccine is delivered via intradermal or intramuscular injection, often using electroporation with Inovio’s CELLECTRA device. Clinical studies have shown that INO-4201 induces strong humoral and cellular immune responses, including robust neutralizing antibody titers and T cell responses characterized by Th1 cytokine production (IFNγ, IL2, TNFa) from both CD4+ and CD8+ T cells. The vaccine has demonstrated safety and tolerability in human trials, with no significant systemic adverse effects reported. Notably, it remains stable at room temperature for over a year—a key advantage for deployment in outbreak settings[2][3][6][9].
