Overview

INO-4600 is a synthetic DNA (synDNA) vaccine candidate developed by Inovio Pharmaceuticals in collaboration with the U.S. Army Medical Research Institute of Infectious Diseases (USAMRIID) for the prevention of smallpox (Variola virus). Utilizing Inovio's proprietary SynCon technology, the vaccine consists of DNA plasmids designed to encode consensus sequences of key viral antigens, specifically L1, A27, B5, and A33. Upon administration via intramuscular injection followed by electroporation using the CELLECTRA delivery system, the plasmids enter host cells and express the targeted antigens. This process elicits a robust immune response characterized by both neutralizing antibodies and T-cell mediated immunity. INO-4600 is designed as a non-replicating, stable alternative to traditional live-attenuated smallpox vaccines, offering a potentially safer profile for immunocompromised individuals and improved logistics for biodefense stockpiling due to its thermal stability.
