INO-4800 is a DNA vaccine candidate developed for immunization against COVID-19. It contains the plasmid pGX9501 encoding the full-length Spike (S) glycoprotein of SARS-CoV-2. The vaccine uses Inovio's proprietary CELLECTRA® device, which delivers brief electrical pulses to open pores in cells, allowing plasmid DNA entry and expression of the spike protein to stimulate both humoral and T cell-mediated immune responses. Preclinical studies demonstrated robust neutralizing antibodies and T cell responses capable of inhibiting viral binding to the ACE2 receptor, critical for viral entry. Clinical trials have shown it to be generally safe, well-tolerated, and immunogenic across age groups. The vaccine is stable at room temperature for over a year without requiring frozen storage or transport, facilitating mass immunization efforts. Development includes Phase 1 through Phase 3 clinical trials globally with ongoing evaluation as a heterologous booster by partners such as Advaccine in China.
