INO-6145 is a synthetic consensus (SynCon®) DNA plasmid vaccine developed by Inovio Pharmaceuticals for the treatment of HIV-1 infection. It is one of two plasmid components (alongside INO-6112) that constitute the PENNVAX-GP vaccine regimen. INO-6145 utilizes codon-optimized DNA sequences to encode HIV-1 antigens, aiming to elicit robust, broad-based CD8+ T cell responses targeting conserved viral epitopes for potential HIV remission. The vaccine is typically co-administered with the DNA adjuvant INO-9012, which encodes interleukin-12 (IL-12), and is delivered via electroporation using the CELLECTRA® device to enhance cellular uptake and immunogenicity. It has reached Phase 2 clinical development for HIV-1 infections.
