Overview

Inobrodib + teclistamab is a combination therapy being evaluated for the treatment of advanced hematological malignancies, primarily relapsed/refractory multiple myeloma. Inobrodib (CCS1477), developed by CellCentric, is a first-in-class, orally available small molecule inhibitor of the p300/CBP bromodomain. It selectively binds to the twin regulatory proteins p300 and CBP, which are transcriptional co-activators that drive the expression of key oncogenes such as MYC and IRF4. Teclistamab (Tecvayli) is a bispecific monoclonal antibody that acts as a T-cell engager, simultaneously targeting the B-cell maturation antigen (BCMA) on malignant plasma cells and the CD3 receptor on T cells to induce directed T-cell-mediated cytotoxicity. This combination is being investigated in Phase I/IIa clinical trials (such as NCT04068597) to explore whether the epigenetic modulation provided by inobrodib can enhance the efficacy or overcome resistance to BCMA-targeted immunotherapy in patients with heavily pretreated multiple myeloma.
