Drug intelligence / Profile preview
Inorganic polyphosphate (polyP) is a naturally occurring, universally conserved linear biopolymer composed of orthophosphate residues linked by high-energy phosphoanhydride bonds. It is stored and released by activated platelets and mast cells, playing a crucial role in hemostasis, bone mineralization, and tissue regeneration. As a therapeutic agent, inorganic polyphosphate is being developed in various formulations (such as sodium polyphosphate and calcium polyphosphate nanoparticles) for applications in bone fracture healing, chronic wound healing, atopic dermatitis, and ulcerative colitis. Its primary mechanism of action involves serving as a concentrated phosphate reservoir for hydroxyapatite formation during bone mineralization, and undergoing enzymatic cleavage by alkaline phosphatase (ALP) and adenylate kinase (ADK) to generate metabolic energy in the form of adenosine triphosphate (ATP), which drives cellular migration, proliferation, and tissue repair.
