Overview

iNOS shRNA is an experimental RNA interference (RNAi) agent designed to silence the expression of inducible nitric oxide synthase (iNOS, also known as NOS2). In the context of oncology research, specifically bladder cancer, iNOS shRNA is used to investigate the role of endogenous nitric oxide (NO) generation in the maintenance and self-renewal of cancer stem cells (CSCs). By targeting and degrading iNOS mRNA, the shRNA reduces iNOS protein levels and subsequent NO production. Preclinical studies in murine models have demonstrated that silencing iNOS leads to a decrease in sphere-forming efficiency and the downregulation of stemness-related genes such as Sox2, Oct4, and Nanog, thereby inhibiting tumor growth and recurrence. This approach is currently utilized as a research tool to validate iNOS as a potential therapeutic target for eradicating CSC populations responsible for disease relapse.
