This is a combination regimen used in clinical studies for the treatment of relapsed or refractory B-cell non-Hodgkin lymphoma, including follicular lymphoma and diffuse large B-cell lymphoma. It pairs two antibody-based therapies: - **Inotuzumab ozogamicin**: an antibody-drug conjugate (ADC) that targets CD22 on B-cells. It consists of a humanized IgG4 monoclonal antibody linked via an acid-labile linker to calicheamicin, a cytotoxic antibiotic. Binding to CD22 leads to internalization, lysosomal cleavage of the linker, and intracellular release of calicheamicin, resulting in double-strand DNA breaks and apoptotic cell death. - **Rituximab**: a chimeric monoclonal antibody that targets CD20 on B-cells, mediating cell lysis via antibody-dependent cellular cytotoxicity and complement-dependent cytotoxicity. These agents employ non-overlapping mechanisms, targeting different antigens on B-cells to provide complementary and potentially synergistic anti-tumor effects. This combination has shown high response rates and manageable toxicity in trials for relapsed/refractory B-cell non-Hodgkin lymphoma[1][2][3].
