Overview

This is a multi-agent chemotherapy regimen combining five drugs: **Inotuzumab ozogamicin** is an antibody-drug conjugate targeting CD22 on B cells. It consists of a humanized IgG4 monoclonal antibody linked to the cytotoxic agent calicheamicin. Upon binding to CD22-expressing tumor cells, it is internalized and releases calicheamicin intracellularly, causing double-strand DNA breaks and cell death[3][6]. **Rituximab** is a monoclonal antibody targeting CD20 on B lymphocytes, leading to their depletion. **Gemcitabine** and **cisplatin** are cytotoxic chemotherapeutic agents that interfere with DNA synthesis and function. **Dexamethasone** is a synthetic glucocorticoid used for its anti-inflammatory and immunosuppressive effects as well as for reducing chemotherapy side effects[7]. This combination has been studied primarily for relapsed or refractory CD22-positive B-cell non-Hodgkin lymphoma—including diffuse large B-cell lymphoma, follicular lymphoma, and mantle cell lymphoma—where it demonstrated promising response rates but also significant hematologic toxicity[1][5].
