Drug intelligence / Profile preview
INP2 is a novel anti-prolyl hydroxylase 2 (PHD2) cytosolic intrabody designed to protect against acetaminophen-induced liver injury (AILI). Developed by researchers at Jilin University, INP2 is synthesized from a human anti-PHD2 single-chain fragment variable (scFv) screened from a phage display library. It specifically binds to PHD2 in the cytoplasm, disrupting the interaction between PHD2 and hypoxia-inducible factor-1α (HIF-1α). This disruption inhibits PHD2-mediated hydroxylation and subsequent degradation of HIF-1α, thereby enhancing HIF-1α protein stability. In preclinical models, INP2 upregulates the HIF-1α/GPX1 axis, restoring glutathione biosynthesis (via upregulation of SLC7A11 and GPX1), improving redox homeostasis, and attenuating ferroptosis and oxidative stress in hepatocytes.
