**INS-3001** is a novel small molecule inhibitor of pathological cardiovascular calcification derived from chemical modifications of myo-inositol hexaphosphate (IP6). It potently stabilizes calciprotein particles, disrupts nucleation and growth of calcified deposits independent of etiology, and demonstrates superior efficacy, pharmacokinetics, and tolerability compared to IP6 in preclinical models of soft tissue calcification, including arterial walls and cardiac valves. Developed by Inositec (a spin-off from ETH Zurich and University of Bern), it targets high-unmet-need indications like aortic valve stenosis (AVS) and peripheral artery disease (PAD), with potential for once-daily subcutaneous administration in ambulatory settings; a Phase 1 trial in healthy volunteers was approved and dosed in late 2021, though no further development reported as of early 2025.[1][2][3][8][9]
