Drug intelligence / Profile preview
INS-406 is a preclinical, hepato-preferential acylated insulin analog developed by Novo Nordisk in collaboration with Vanderbilt University. It was engineered to distribute more like portal insulin when delivered subcutaneously, thereby mimicking the physiological portal-to-peripheral insulin gradient. By preferentially targeting the liver over peripheral tissues (such as muscle and fat), INS-406 aims to restore normal hepatic glucose metabolism and suppress hepatic glucose production while minimizing peripheral glucose utilization. This hepato-preferential distribution profile is designed to provide a protective effect against hypoglycemia compared to conventional peripheral insulin therapies, making it a promising candidate for the treatment of diabetes mellitus.
