Drug intelligence / Profile preview
Insparin is a recombinant protein derived from the E4orf1 gene of human adenovirus Ad36 (HAdV-D36). It is being developed as a first-in-class antidiabetic agent that promotes cellular glucose uptake in an insulin-independent manner. Insparin bypasses the proximal insulin signaling pathway (insulin receptor and insulin receptor substrate) and directly activates the distal Ras-PI3K-AKT pathway. It binds to Drosophila disk large protein (DLG1) via its PDZ-binding motif to trigger this signaling cascade. Preclinical studies have shown that subcutaneous administration of Insparin encapsulated in soy-phosphatidylcholine liposome nanoparticles improves glycemic control, reduces HbA1c, and promotes fat loss and liver fat reduction without causing hypoglycemia.
