InstanCAR-T is a next-day, CD19-targeted CAR-T cell therapy product designed for rapid manufacturing, typically within two days, significantly faster than conventional CAR-T therapies. This rapid production aims to preserve a less-differentiated T-cell phenotype, enriching for naive and stem cell memory T cells, which contributes to enhanced proliferation kinetics, greater CAR expression density, and improved persistence. It demonstrates potent anti-leukemic activity with a favorable safety and efficacy profile, as evidenced by durable tumor control in murine xenograft models and comparable clinical outcomes to conventional CAR-T in a Phase I trial for relapsed/refractory B-cell acute lymphoblastic leukemia (R/R B-ALL). High-dimensional immune profiling indicates sustained enrichment of stem-like T-cell subsets and reduced exhaustion markers, supporting its potential for robust and durable therapeutic effects.
