Drug intelligence / Profile preview
Insulin B:9-23 peptide mimotope nanobubbles is an image-guided, submicron-sized lipid-shelled nanobubble (NB) formulation encapsulating a palmitoylated insulin B:9-23 peptide mimotope. Developed as a theranostic platform for type 1 diabetes (T1D), these nanobubbles act as ultrasound contrast agents that passively accumulate in pancreatic islets due to the increased microvascular permeability associated with insulitis. Once localized, they can be visualized and noninvasively burst using contrast-enhanced ultrasound (CEUS) to deliver the tolerogenic peptide cargo. This targeted delivery promotes the expansion of anti-inflammatory, insulin-reactive CD4+CD25+Foxp3+ regulatory T cells (Tregs) in the islet microenvironment, thereby inducing peripheral immune tolerance and delaying or preventing the autoimmune destruction of insulin-producing beta cells.
