Insulin glargine U300 is a long-acting human insulin analog formulated at a concentration of 300 units/mL. It is used to improve glycemic control in adults with type 1 and type 2 diabetes mellitus. Insulin glargine mimics endogenous human insulin by promoting glucose uptake into muscle and fat tissue and inhibiting hepatic glucose production. The higher concentration (U-300) provides a more prolonged and stable release profile compared to the standard U-100 formulation, resulting in up to 36 hours of action with a delayed onset (up to 6 hours). This allows for once-daily dosing with reduced risk of hypoglycemia between meals or overnight. Insulin glargine also inhibits lipolysis and proteolysis while enhancing protein synthesis[1][3][6][9][10].
