The insulin-like growth factor 1 receptor (IGF1R) vaccine is an investigational therapeutic protein subunit vaccine candidate designed to target the IGF1R signaling pathway in cancer. Developed by CK Life Sciences, the vaccine utilizes recombinant proteins that incorporate computationally-predicted B-cell epitopes from the IGF1R extracellular domain, specifically targeting regions involved in the interaction between the receptor and its ligands (IGF1 and IGF2). The vaccine is formulated with immune enhancers and adjuvants, such as CpG ODN 1826 and AS03, to induce high titers of neutralizing antibodies. These antibodies competitively block the binding of IGF ligands to IGF1R, thereby inhibiting downstream signaling pathways that promote tumor cell proliferation, survival, and resistance to conventional therapies. Preclinical studies in syngeneic mouse colon cancer models (MC38) have demonstrated that the vaccine can elicit robust humoral immune responses and significant anti-tumor activity, suppressing tumor growth by over 60%.
