Overview

Insulin-like growth factor 1 (IGF-1) small interfering RNA is a therapeutic modality designed to silence the expression of the IGF-1 gene through the RNA interference (RNAi) pathway. By targeting the mRNA transcript of IGF-1, these siRNAs trigger its degradation, thereby reducing the production of the IGF-1 protein. IGF-1 is a potent growth factor involved in cell proliferation, survival, and transformation, and its dysregulation is implicated in various cancers (such as acute myeloid leukemia, prostate, and breast cancer) as well as growth disorders like acromegaly. In the context of oncology, IGF-1 signaling often promotes constitutive activation of the PI3K/Akt pathway, contributing to tumor growth and resistance to therapy.
